Overview

Ipamorelin is a synthetic peptide composed of five amino acids, classified as a Growth Hormone Secretagogue (GHS). Its structure is Aib-His-D-2-Nal-D-Phe-Lys-NH2. Ipamorelin was designed to bind to ghrelin receptors (GHS-R) on the pituitary gland, which regulate natural growth hormone (hGH) secretion. This peptide is recognized for being one of the first highly selective synthetic GHS compounds.

Research Background

Ipamorelin is studied for its potential to selectively stimulate growth hormone release without significantly affecting other pituitary hormones such as prolactin or ACTH. By promoting hGH secretion, Ipamorelin may indirectly increase IGF-1 production, which mediates many of hGH’s anabolic actions, including tissue growth, cellular repair, and bone and muscle metabolism.

Mechanism of Interest

  • Selective GH release: Binds to GHS-Rs on pituitary cells, potentially stimulating growth hormone secretion.
  • Anabolic activity: May promote cellular proliferation, bone density support, and muscle anabolism via IGF-1 signaling.
  • Lipolysis: Suggested to influence fat metabolism through growth hormone–mediated pathways.
  • Specificity: Reported to avoid stimulation of other pituitary hormones such as prolactin and ACTH, distinguishing it from earlier GHS compounds.
